Membrane proteins are embedded in cell membrane and have the role of connecting intracellular and extracellular environments. Rapid and quantitative analysis of membrane proteins have been strongly required in drug discovery research. These days, Liquid chromatography-mass spectrometry (LC/MS) has widely been used for analyzing proteins. However, it is difficult for LC/MS to analyze membrane proteins because detergents that are used sample preparation suppress ionization by electrospray ionization (ESI) that are used as an ionization method. In this study, an atmospheric pressure infrared matrix-assisted laser desorption electrospray ionization (MALDESI) was developed. This is the combined method of ESI and an atmospheric pressure infrared matrix-assisted laser desorption ionization (MALDI) that is hardly effected by detergents and can be connected LC directly. As a result, compared with atmospheric infrared laser ionization (without spray), Ion signal intensity of angiotensin II (peptide) was 2 times increased with this method (with spray). This method can be expected for membrane proteins by investigation of the ion signal intensity when measuring proteins.
