糖タンパク質解析のためのメソポーラス有機シリカ薄膜基板を用いたレーザー脱離イオン化質量分析

Matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) is powerful for the analysis of glycans and glycopeptides due to soft ionization. Inhomogeneneous crystallization of analytes with a matrix, 2,5-dihydroxybenzoic acid (DHBA) causes some problems regarding quantitation and reproducibility. We developed a new substrate made of periodic mesoporous organosilica films, which can absorb N2 laser. Without a conventional matrix, a sub-picomol of a dye-labeled glycan was directly detected by laser irradiation using MALDI-TOF MS. The signal production needs the laser-absorbing functional group in the films and the presence of mesopores extremely enhances signal intensity. Some results suggest that excitation energy transfer from the organosilica framework to the dye contributes to success in laser desorption/ionization.
