Improving LC-MS/MS Bioanalytical Quantitation of Oligonucleotides Using New High Performance Tandem Quadrupole Mass Spectrometer

Improving LC-MS/MS bioanalytical performance especially for ESI negative analytes has been of high interest especially for the challenging analysis of ONTs (oligonucleotide therapeutics) in biological matrices. Developing low/sub ng/mL Oligonucleotide therapeutic quantitation methods requires special attention to recovery, removal of protein/interference and losses due to non-specific binding. This work will discuss the capabilities of a new high performance tandem quadrupole mass spectrometer in combination of high-performance surface treated consumables and LC-systems to improve detection limits. The strategy will be demonstrated through quantification of Oligodeoxythymidines and the fully phosphorothioated antisense oligonucleotide therapeutic, GEM91 (Trecovirsen).
In particular, TQ enhancements focused on the improvement of negative ion mode sensitivity performance will be discussed. Incorporating sound preparative approaches with the latest LC and MS technologies enables routine low ng/mL quantitation of ONTs.
