Nucleic acid analysis using MALDI-DIT-MS

Recently, practical application of nucleic acid medicines has rapidly advanced, and the importance of nucleic acid analytical technique has increased. However, reports of nucleic acid analysis by matrix-assisted laser desorption/ionization (MALDI) method have been limited. In this study, we attempted to develop a nucleic acid analysis method using MALDI-digital ion trap-mass spectrometer (MALDI-DIT-MS). Nucleic acid of MW 6201 was detected with high sensitivity as [M+H]+ using 3-hydroxypicolinic acid as a matrix and ammonium citrate as an additive. In-source decay (ISD) of the nucleic acid was improved by optimizing equipment condition and matrix. Sequencing analysis was accelerated by in-house software. Finally, rapid molecular weight analysis and sequence analysis including modification site were achieved for nucleic acid medicines after desalting purification.
