Development of LC-MS/MS based comprehensive analytical method for steroidogenesis assay using human adrenal cortex cancer cell line

Steroid hormones such as 17β-estradiol (E2) and aldosterone secreted from reproductive-related tissues and the adrenal glands play an extremely important role in development of reproductive organs and maintaining homeostasis. Changes due to endocrine effects of the new drug candidate are often observed in toxicity studies. The steroid hormone assay system using human adrenal gland cell line described in OECD test guidance 456 is useful evaluation system for endocrine disruption effect of test chemicals. This assay is target only testosterone and E2, and we tried to expand targets using LC-ESI-MS/MS based assay system. Sixteen steroid hormones in cell culture medium could be analyzed comprehensively and quantitatively in combination with derivatization. The sensitivity of this analytical method reached to pg/mL order in cell culture medium. In addition, steroidogenesis evaluation for 5 different inhibitors was successfully evaluated by using this assay system.
