Improvement of sensitivity for the LC/MS analysis of GSH and its metabolites by using Bio LC system

Glutathione (GSH) and its metabolites play an important role in oxidoreduction homeostasis and oxidative stress tolerance1). Glutathione and its metabolites are a group of compounds with thiol groups. Due to their reactivity with, it is difficult to obtain a good peak shape by general HPLC system. So, thiol groups are often specifically protected with… to obtain a good peak shape or in some cases fluorescently labeled for highly sensitive analysis2). However, derivatization process is time-consuming, and there are concerns that large excesses of labeling reagents may desensitize the mass spectrometer. In this study, we develop a method for quantitative analysis of underivatized GSH and its metabolites using an Agilent 1290 Infinity II Bio LC with MP35N® material. MP35N® material has excellent chemical resistance, then can be used over a wide pH range and salt concentration. Therefore, it is expected to be more inert to thiol groups than stainless steel (SUS), which is commonly used in general LC.
