Enhanced sensitivity of DIUTHAME, a matrix-free laser desorption / ionization method

We developed DIUTHAME (Desorption Ionization Using Through Hole Alumina MEmbrane) which enables measurements of MALDI without matrix compounds. Output signal from DIUTHAME is sometimes lower than that of MALDI, partly because of its low absorption of laser energy. This study aims to improve the absorption efficiency of laser energy in order to increase the signal intensity of DIUTHAME. As improvement method, the through-holes of DIUTHAME were developed into a tapered shape, and metal nanoparticles were formed on the Pt conductive layer of DIUTHAME surface. From the results of MS imaging of mouse brain sections, it was confirmed that the average signal intensity of phospholipids was improved by 10 times.
