Tapping-mode scanning probe electrospray ionization (t-SPESI) is a extraction-ionization technique using picoliter solvents, which can be applied to mass spectrometry imaging (MSI). Positive ion mode MSI of mouse brain tissue sections was studied using DMF/MeOH binary mixtures and their respective pure solvents. The mixed solvent was found to have the highest number of ion peaks detected, and the physicochemical properties of the solvent were found to influence the extraction and ionization in t-SPESI.
