Peptide sequence analysis using MS2 of alkali metal ion adducts

In matrix-assisted laser desorption/ionization (MALDI), peptides are often detected as [M+H]+, but they can also be observed as alkali metal adducts, such as [M+Na]+. In this study, MS2 of alkali metal ion adducts ([M+Li]+, [M+Na]+, [M+K]+, [M+Rb]+ and [M+Cs]+) and [M+H]+ for peptides were compared. As a result, MS2 fragments for all the alkali metal ion adducts were mainly detected as y-ions or b-ions. In particular, ratio of total peak intensity of y-ions to total peak intensity of b-ions was highest in the [M+Cs]+ MS2 spectrum.
