A comprehensive method for simultaneous determination of bioactive lipids by LC/MS/MS

The comprehensive measurement of blood metabolites is expected to be used for early detection and risk assessment of diseases. However, most of the cohort studies to date have not been clinically applied. One reason for this may be that primary metabolites, which have been the target of many studies, vary with various environmental factors and thus have low disease specificity. Bioactive lipids such as sterols and eicosanoids bind to a nuclear receptor and membrane receptor and exert physiological and pathophysiological activity in biological systems. Therefore, comprehensive and quantitative information on bioactive lipids in the blood is expected to provide useful indicators for a better understanding of health status. Here, we have developed a new analytical method that combines solid phase extraction (SPE) and a liquid chromatography tandem mass spectrometry (LC/MS/MS) to observe a wide range of over 150 bioactive lipids. Our developed method detected over 70 bioactive lipids in human plasma of clinical specimen or normal healthy volunteers (NHVs). We evaluated the gender differences, inter-day variability, and intra-day variability in the blood of NHVs. We hope to find new interesting collaborations using this method through discussions.
