Peptidomics based on high-efficiency peptide extraction method.
The differential solubilization (DS) method that we developed enables high-efficiency extraction of peptides from serum/plasma, including peptides bound to carrier proteins such as albumin. Using this method, we directly identified an authentic peptide hormone using LC-MS and various novel bioactive peptides. Furthermore, we modified the DS method to make it applicable to tissue samples. The modified DS method achieved highly efficient and reproducible peptide extraction from a few milligrams of frozen tissue. Subsequent LC-MS/MS analysis successfully resulted in the identification of numerous authentic bioactive peptides and a novel bioactive peptide from animal tissues. The method based on our peptide extraction technique may facilitate the discovery of novel bioactive peptides and peptides that are related to disease.
