Extracellular vesicles (EVs) from the cells are facilitators of cell-to-cell communication. EVs released by cancer cells are implicated in cancer progressions, including distant metastasis, angiogenesis, and immunosuppression. EVs include miRNAs, mRNAs, DNA, proteins, and lipids, which have been intensively studied, however, metabolites remain still poorly understood. In this study, we established a system for comprehensive metabolomic analysis of EVs. EVs in culture supernatant of human pancreatic cancer PANC1 cells were collected by ultracentrifugation and analyzed for hydrophilic metabolites by capillary ion chromatography/mass spectrometry (capIC/MS) and liquid chromatography/mass spectrometry (LC/MS) and lipids by supercritical fluid chromatography/tandem mass spectrometry (SFC/QqQMS). We identified 140 hydrophilic metabolites and 494 lipids in pancreatic cancer-derived EVs, and their profiles were different from those in cells. We also found that the levels of hydrophilic metabolites in EVs were changed by hypoxic stress. Currently, we are developing a new method for EVs collection using size exclusion chromatography to improve the collection efficiency. In this presentation, the current status will be introduced.
