血液メタボロミクスデータ統合に向けたLC/MS分析法の定量性評価
Metabolomics, mass spectrometry-based comprehensive analysis of metabolome, has attracted attention as an analytical approach to provide novel insight into metabolism and to search for biomarkers. However, one of the current problems in the field of metabolomics is the difficulty in integrating data collected using different equipment at different facilities, because many metabolomic methods have been developed independently and are unique to each laboratory. Herein, we evaluated the quantitative performance of four types of liquid chromatography (LC), including hydrophilic-interaction LC (HILIC), ion chromatography with an anion exchange column (AEX), LC with pentafluorophenylpropyl column (PFPP-LC), and unified HILIC/AEX, coupled with benchtop quadrupole Orbitrap high-resolution mass spectrometry for hydrophilic metabolome analysis of human plasma.
