Steroid profiling by LC-MS/MS: General linear model for prediction of biological age

Aging is defined as the process of accumulation of damage in individuals from molecular to cellular level, systemic decline in organ function, and the development of disease, which eventually leads to the death of the individual. Aging is associated with genes, environment and lifestyle, with genetic factors accounting for only 20-25%, and non-genetic factors contributing to a much greater extent to human aging. As the average human lifespan increases, various age-related diseases are becoming apparent. Therefore, it is important to find an accurate way to determine biological age which reflects the physiological and mental condition of the individual, to have information on potential biomarkers and to understand their specific relationship with aging for the diagnosis and even prognosis of diseases.
In this study, we collected blood samples from people aged 20-70 years and quantified 29 steroid hormones in their serum by using LC-MS/MS detection technique developed by ourselves. By performing multivariate analysis of the quantitative results, we have found potential biomarkers related to age. The quantitative data of these potential biomarkers were used to build and test a generalized linear model and to compute a fitting curve for biological age.
