アミノ酸バイオマーカー測定法開発と応用
Amino acids are known as one of the important metabolites in living organisms, and their concentrations are strictly controlled by homeostatic regulation. The changes of amino acid concentrations in various diseases were reported. We have developed analytical method for amino acids using pre-column derivatization reagents (APDSTAG®) combined with LC-MS/MS and applied the method to biomarker research. Using the analytical method, we found the slight changes of amino acid concentration balance in various cancer and life-style disease and developed “Aminoindex® risk screening” test. For highly accurate amino acid analysis using a mass spectrometer, the quantification range was narrowed down according to the plasma concentration balance, and stable isotope labeled amino acids were used as an internal standard to correct the matrix effect. Furthermore, analytical method for amino acid metabolites and chiral amino acids were also developed. Especially in urine and feces, the ratio of D-amino acid is relatively high, and it is necessary to determine D-amino acids and L-amino acids separately. We originally designed pre-column derivatization reagent (R)-BiAC for chiral amino acid analysis and applied for urine and feces biomarker research.
