酸素添加酵素が産み出す多彩な生理活性分子のin situイメージング

Low-molecular-weight cellular signaling mediators, such as monoamines, prostaglandins and steroid hormones, orchestrate a programmed progression of a series of biological phenomena, including inflammation. It is worth to note that all those molecules are produced by oxygenase activity. Therefore, in order to understand the biochemical basis of inflammatory diseases, it is important to understand the spatiotemporal dynamics (i.e., production, diffusion, and decomposition) of such signaling mediators in tissues; however, the lack of effective molecular imaging technology has made it difficult to determine their localizations in vivo. We have attempted to fill this technical gap by developing highly sensitive imaging mass spectrometry (IMS). Here, I will report recent progress in this field and how IMS has contributed to understand metabolic activity of specific types of immune cells, even inside lymph nodes tissues. Moreover, stable isotope labeling techniques utilizing 13C, 15N and 18O2 allowed us to visualize flows of specific metabolic pathways in tissues. I’ll also show that how the upregulated immune metabolism alters brain monoamine signaling, by visualization of deficiency of the neurotransmitter serotonin, which finally resulted in abnormal animal behavior.
