タッピングモード走査型プローブエレクトロスプレーイオン化法によるヒト拡張型心筋症組織の質量分析イメージング
Mass spectrometry imaging (MSI) is a technique to measure the multidimensional chemical information to visualize the distribution of components in samples. We have been developing tapping-mode scanning probe electrospray ionization (t-SPESI) and applying it to MSI of biological tissues. In this presentation, we will report the results of high mass resolution MSI of human diseased heart tissues using a t-SPESI system with a built-in feedback control device. The MSI results showed that there were multiple lipids whose ion intensities were distributed throughout the tissue, as well as lipids localized within the tissue. The localized lipids were tentatively assigned as triacylglycerols (TG). The localization information of TG species in cardiac tissues may lead to the stratification of pathophysiology and the prognosis prediction of dilated cardiomyopathy. We showed that MSI by t-SPESI to measure the distribution of multiple TGs, which is difficult to achieve using staining methods.
