Py-Tag 誘導体化試薬によるパーキソンモデルラット脳内の解析

To demonstrate a faithful analysis of catechol amine and amino acid with
derivatization reagent, we investigated reaction condition of Py-Tag as
derivatization reagent with target dopamine and GABA on section and
constructed optimized reaction holder. Ten different reaction conditions
of Py-Tag with target were considered. As an optimal condition, Py-Tag
was able to react with target in 70% methanol with 5% trimethylamine
(v/v) solution at 60 ℃ in homogenous condition. To reproduce this
reaction on section, we made the reaction holder that kept moisture
condition and facilitate derivatization reaction. Especially,
visualization of DA and GABA was archived by derivatized-imaging mass
spectrometry (IMS). A brain section of unilateral 6-OHDA lesioned
Parkinson’s disease model rat showed Py-Tag dopamine (m/z 328.3) at
unilateral striatum and Py-Tag GABA (m/z 278.3) at cerebral cortex,
striatum, hippocampus and hypothalamus. By using PD model rat brain, we
obtained images with left-right difference for the localization of DA
and GABA.
These findings suggest that the consideration is important in the
reaction condition that allows DA or GABA and Py-Tag to react with high
efficiently and to construct high quality image on section.
