微量試料測定に向けた高速プロテオーム解析システムの高感度化検討
Shotgun proteomics using nano liquid chromatography/tandem mass spectrometry typically involves low flow rates of less than 1 µL/min. However, such conditions require a long time for column washing and equilibration, which is a major obstacle when performing high throughput measurements. Therefore, we constructed a high-speed LC/IMS/MS system using capillary LC, and succeeded in reducing the measurement time from 2 hours to 2 minutes. On the other hand, the use of capillary LC at several µL/min reduces sensitivity compared to nano LC, which may make it difficult to apply to minute samples. In this study, we measured minute samples using the high throughput capillary LC/IMS/MS/MS system and found that an average of 2,235 protein groups were identified for a 250 ng sample, whereas 111 protein groups were identified for a 0.5 ng sample. In this presentation, results using a hydrophilic interaction chromatography (HILIC) column will also be reported.
