星細胞腫と乏突起膠腫の術中鑑別診断に利用可能なバイオマーカーの探索

Astrocytoma and oligodendroglioma are brain tumors classified as gliomas. Because very little residual astrocytoma leads to recurrence and intraoperative differential diagnosis is required to aim for aggressive surgical removed. Although the characteristics of genetic mutations and chromosomal aberrations in both tumors have been reported, there is no molecular diagnostic methods that can be quickly used and differentiate them. Therefore, we aimed to search for biomarker candidates for differentiating them with metabolomic analysis using mass spectrometry and develop a molecular diagnostic method based on quantitative analysis.
First, we searched for peaks that increased in two tumors by global metabolomics and speculated biomarker candidates with metabolomic database, and identified them using authentic standards. Next, we quantified the biomarker candidates in brain tumors and explored models for differentiation.
As a result, hypoxanthine, inosine, and cystine were detected at high levels in tumors of oligodendroglioma patients against astrocytoma. The diagnostic model generated by the decision tree method with the concentrations, showed high discriminative performance. It was suggested that their mass spectrometric analysis in surgery and the diagnostic model might be useful for differentiating astrocytoma and oligodendroglioma.
