大気圧連続流赤外レーザーエレクトロスプレーイオン化法を用いた緩衝液または塩を含んだぺプチド試料の測定およびESI法との比較
Liquid chromatography/mass spectrometry (LC/MS) has been widely used for analyzing biomolecules in drug discovery research. However, buffer or salts in sample preparation largely decrease ionization efficiency in conventionally used electrospray ionization (ESI). In this study, infrared (IR) laser desorption electrospray ionization has been developed for proteins. Solutions of a peptide, angiotensin II, containing a buffer, Tris-HCl or salt, NaCl, were measured using this method or ESI, and ion signal intensities of the peptide were compared. As the results, it became clear that IR laser desorption electrospray ionization has higher tolerance to Tris-HCl and NaCl compared to ESI.
