Liquid chromatograph-mass spectrometers (LC-MS/MS) are used for variety of application as a highly selective and sensitive quantification technology. In some cases, the reproducibility, sensitivity, and robustness of the measurement systems (sample preparation, LC separation and MS detection) are affected by the physicochemical properties of the analyte and/or matrices, and it takes a longer time to develop a LC-MS/MS method. In particular, phosphoric acid, carboxylic acid, or amino functional groups will interact with metal surface of analytical systems, then cause poor peak shape and resulting in decreased sensitivity, reproducibility, and robustness. This work will discuss solution for challenging LC-MS/MS method development for perfluoroalkyl substances (PFAS) in drinking water and oligonucleotides bioanalysis as examples.
