Development of Chiral Stationary Phase for Chromatography Using Mass Spectrometric Screening

As inulin which is one of polysaccharides has polyoxyethylene structure as the main chain, it makes noncovalent complex with cationic species via charge-dipole electrostatic intermolecular interaction. The chiral discrimination ability of the oligomer mixture of inulin derivative (methyl, benzyl, acetyl, and benzoyl) toward several 2-propyl ester of protonated chiral amino acid was evaluated using fast atom bombardment mass spectrometry coupled with the isotopically labeled guest method which was proposed by Sawada et al. Permethylated inulin among the inulin derivatives showed high chiral discrimination ability toward given chiral cations. Permethylated inulin was fixed on silica gel to use as chiral stationary phase in liquid chromatography and the chiral separation factor toward chiral ammonium ion was examined. The separation factors values showed linear relationship to the chiral discrimination ability in the FABMS screening.
