Development of siRNA analysis method using MALDI-DIT-MS

Practical application of oligonucleotide therapeutics has rapidly advanced, and the importance of nucleic acid analytical technique has increased. Herein, we report siRNA analysis method using matrix-assisted laser desorption/ionization-digital ion trap-mass spectrometer (MALDI-DIT-MS). Vutrisiran was detected mainly as two peaks in the sense and antisense chains. By adjusting the instrument setting conditions for high-mass analysis, [M+H]+ as a double strand was also observed. By adjusting the setting conditions for sequencing analysis, many fragment ions were detected and the sequence of the sense and antisense chains was analyzed. At this time, three peaks corresponding to the terminal triantennary GalNAc modifications was also detected, and MS2 and MS3 of them provided structural information of the triantennary GalNAc modifications. We propose a simple qualitative analysis method for siRNA using MALDI-DIT-MS.
