Application of Targeted Proteomics by nano-LC-MS/MS to Lipomyces starkeyi

Targeted proteomics using nano-LC-MS/MS was applied to the oleaginous yeast Lipomyces starkeyi. The protein extraction method was optimized by using a multi beads homogenizer. MRM assay methods including 2,288 transitions were also constructed to measure the expression levels of 107 enzyme proteins. The targeted proteomic analysis of the central metabolism-related enzymes revealed that those expression levels in the lipid production phase were significantly lower than that of the exponential growth phase.
