Development of Amyloid β Certified Reference Material

Amyloid β is known as one of the biochemical biomarkers of Alzheimer's disease. We have developed amyloid β certified reference material, NMIJ CRM 6210a. This CRM is the lyophilized synthetic peptide having a human amyloid β (1-42) sequence (thereafter, amyloid β (1-42) is defined as amyloid β). The certified values of this CRM were the mass concentration of amyloid β and total amyloid β, determined via amino acid analysis employing an isotope-dilution mass spectrometry for the constituted solution of the candidate material with 1.00 g of 0.1 % ammonia solution. The results of amino acid analyses were converted into the mass concentration by using the density and molecular weight of the reconstituted solution to give the certified values. The content of amyloid β in total amyloid β was determined by the relative area percentage using HPLC. The peptide impurities were identified by high-resolution mass spectrometry.
