The practice of epitope mapping against a glycoprotein by the latest HDX-MS system

Recently, it has been reported that epitope mapping of monoclonal antibody against hemagglutin was performed by Hydrogen-Deuterium Exchange Mass Spectrometry (HDX-MS), while glycosylation sites were uncovered due to the difficulty of detecting and quanitfying peptides containg glycans by HDX-MS1). Here, we will introduce an approach for epitope mapping on glycosylated SARS-CoV2 receptor-binding domain of S protein by the latest HDX-MS system consist of Orbitrap Eclipse, PAL and UPLC.
