バイオ医薬品開発における効率的な糖鎖解析のためのワークフロー
We present software-driven MS-based methods that provide a complete and detailed overview of biopharmaceutical glycosylation at the intact protein, peptide, and released glycan levels. MS data from various instruments and biopharmaceutical samples was processed and analyzed using Genedata Expressionist® (Genedata AG, Basel, Switzerland). Glycoanalysis of intact proteins enabled identification and quantification of glycosylated species in a range of biopharmaceutical preparations. Deconvolution mode and algorithm were adapted to the input data resolution, and time-resolved deconvolution revealed additional heterogeneity. In glycopeptide analysis, expected glycosylation sites and consensus sequences were factored into data processing. The flexibility of glycan libraries searched and adducts considered enabled not only highly targeted analyses, but also dramatically accelerated data analysis by reducing the number of possible matches and consequently the number of false-positive identifications. Analysis and identification of released glycans was supported by consolidation and automatic annotation of MS/MS fragment spectra and custom glycan libraries were used to speed up processing and reduce the number of false positives.
