温度制御逆相クロマトグラフィーによるプロテオーム解析
Successful application of mass spectrometry-based shotgun proteomics is heavily relying on the quality of reversed phase liquid chromatography (RPLC). In RPLC, temperature plays a key role since the majority of the chromatographic properties are a function of temperature. However, the importance of appropriate temperature control of stationary phases is often neglected, both in solid-phase extraction step and LC/MS/MS analysis, which are both critical steps to improve the peptide sensitivity as well as system stability. Here, we developed temperature-controlled proteomics workflow to maximize the identification of both hydrophilic and hydrophobic peptides.
