Matrixフリーレーザー脱離イオン化法:DIUTHAMEを用いた質量分析イメージングの高感度化
We developed DIUTHAME (Desorption Ionization Using Through Hole Alumina MEmbrane) instead of matrix compounds in MALDI. Matrix-free DIUTHAME doesn’t have proton donating function in principle, so that it sometimes lower signal than MALDI. This study aims to examine the signal intensity can be improved by coating the DIUTHAME chip with a cationizing agent. Comparing the MS imaging results of the mouse brain section between with and without NaCl coating on DIUTHAME chip, it was observed that the signal intensity increased at [M+Na]+ of phosphatidylcholine with the NaCl-coated chip. This result was presumably caused by integrating the signals separately detected as protonated, sodiated, and potassiated molecules into sodium cationized one. it was suggested that the DIUTHAME chips coated with the cationizing agents can improve the signal intensity while utilizing the characteristics advantages of DIUTHAME.
