生細胞からサンプリングした試料のネイティブ質量分析
Native mass spectrometry (Native MS) with nanoESI enables observation of intact ions of protein complexes with a small amount of sample. It has long been considered that the samples for native MS are required to purify as much as possible and to prepare in volatile solutions, such as aqueous ammonium acetate. In the present study, we examined native MS to observe intact ions of protein complexes without any purification procedures from the crude cell lysate. Optimization of the experimental conditions allowed detection of stable protein ions. Furthermore, intact hemoglobin ions could be observed for the crude red blood cells, without purification with any chromatography.
