Serotonin is produced in the brain and gut, and it plays important roles in both organs. This suggests serotonin functions in brain-gut interactions. The relationship between serotonin and dysphoria has been studied. The serotonin content in feces can indicate the status of serotonin production in the gut and can be an indicator of dysphoria. Analyzing trace substances in feces is very difficult due to the large amount of contaminants. Therefore, to develop a method of analyzing serotonin content in feces, we used solid phase extraction and column-switching LC/MS with human feces containing stable isotope-labeled serotonin [1]. We analyzed 28 feces donated by 20 subjects, and obtained recovery rates of 58.2±2.5 % (n=6) in one day and 60.8±9.1 % (n=5) over multiple days using one of the feces. The content of endogenous serotonin in the 28 dry feces was 0.30-9.93 ng/mg.
