MALDI-ISDを用いるアミノ酸からの水素引き抜き反応

Hydrogen abstraction is a reaction involving radical transfer and degradation of biomolecules, and this reaction is important for understanding the radical related processes. Here we study the mechanism of hydrogen abstraction from amino acids and peptides using matrix-assisted laser desorption/ionization (MALDI). When 2,4-dinitro-1-naphthol (DNNL) was used, the losses of hydrogen and formic acid from amino acids were observed, but their intensities were dependent on amino acids. The ion peak intensities originated from the loss of hydrogen from methionine and cysteine were larger than that from other amino acids, while hydrogen loss was hardly observed in the amino acids with basic side chains and glycine without β carbon. Also the ion peak intensity originated from the hydrogen loss from dipeptides was dependent on the number of nitro group of matrix. The formation mechanism of a-ion resulted from Cα-C bond cleavage was investigated by using a deuterated amino acid Ala(d3).
