MALDI - イメージング質量分析法を用いた実験的自己免疫性脳脊髄炎マウス中枢神経系組織プロテオーム解析による多発性硬化症のバイオマーカーと治療標的の特定

Multiple sclerosis (MS) is an autoimmune disease its cardinal symptoms are chronic inflammatory, demyelination and axonal degeneration of central neuro system (CNS). In this study, we applied matrix assisted laser desorption/ionization (MALDI) imaging mass spectrometry (IMS) for CNS of murine EAE without on-tissue digestion. We have successfully identified and visualized some specific proteins specifically expressed in lesions and investigated a candidate of therapeutic target. It was validated on an EAE model with or without blocking agents of the proteins. Furthermore, using LC-TIMS-MS/MS, in depth proteomics for the cells infiltrated into CNS was also performed.
