Rapid Molecular and Structural Characterization of Acidic Hydrolyzates from 4-O-Methylglucuronoxylan Generated by Endoxylanases Using Electrospray Multistage Mass Spectrometry with Diagnostic Product Ions
Electrospray mass spectrometry in the negative ion mode enhances the detection of acidic xylo-oligosaccharides released from the enzymatic hydrolysis of 4-O-methylglucuronoxylan to rapidly evaluate the distribution of degradation products. An efficient one-way depolymerization via stepwise glycosidic bond cleavages at the reducing end further allows the unambiguous localization of the acidic moiety along the oligoxylose backbone in multistage mass spectrometry relying on the detection of pairs of diagnostic product ions. It ultimately reveals the mode of action of well-known and new endoxylanases.
