Tapping-mode scanning probe electrospray ionization (t-SPESI) is the ambient sampling ionization (ASI) method utilizing the formation, breakage of nano-volume liquid bridge (NLB) and electrospray ionization at the apex of capillary probe. Previously, we reported the simultaneous measurement of the probe displacement and the ion signal during a single sampling and ionization process for the study of the dynamic change of NLB. t-SPESI is expected to realize high spatial resolution by reducing the volume of NLB. In this presentation, we show the duration control of liquid bridge in SPESI for the reduction of sampling spot. For the simplified setup, the new optical detection method was developed. The laser light is irradiated from the side of a probe to generate the probe shadow. The movement of the shadow is detected by using a quadrant photodiode. We combined the optical detection method and feedback control system for the duration control of liquid bridge. We will present the relationship between the duration time and ASI efficiency.
