LC/MS/MSによる人工多能性幹細胞 (iPSC) 及びiPSC由来細胞の特性解析

Induced pluripotent stem cells (iPSCs) and iPSC-derived cells are useful in regenerative medicine, disease modeling, and drug safety research. However, directed differentiation to target cell types is still challenging, and in-depth characterizations of iPSCs and iPSC-derived cells are required for commercial application. We have conducted LC/MS-based proteomics, phosphoproteomics, and glycoproteomics for elucidation of differentiation mechanisms and characterization of iPSCs and iPSC-derived cells. Using LC/MS/MS, we simultaneously confirmed up-regulations of multiple proteins and pathways which are related to the differentiated cell functions. Moreover, differentiation stage-specific alteration of glycosylation was observed in several glycoproteins. In this study, we revealed LC/MS/MS is a powerful tool for characterization of iPSC-derived cells and elucidation of their differentiation mechanisms.
