脂質由来ラジカル種の構造解析法
Lipid-derived radicals generated by biological oxidants are key molecules in the onset and progression of a variety of diseases. To better understand such diseases, a method for the identification and quantitative detection of lipid-derived radicals is desirable. A high-performance liquid chromatography-fluorometry and high-resolution tandem mass spectrometry system combined with a fluorescent probe,NBD-Pen, was developed and used to analyze the adducts formed between NBD-Pen and the radicals derived from five different polyunsaturated fatty acids after enzymatic and non-enzymatic initiation of peroxidation. A database of 134 lipid-derived radicals, including 112 new species, was constructed and the structures of the radicals could be used to provide information on the type of the initial fatty acid. In addition, 12 endogenous lipid-derived radicals were identified in an animal model of liver disease. This method will be useful for the analysis of endogenous lipid radicals and in research toward better understanding of the mechanisms of related diseases.
