Characterization of Biopharmaceuticals: Development of Disulfide Linkage Analysis Method for Human Hepatocyte Growth Factor
Human hepatocyte growth factor (hHGF) is about 90 kDa heterodimer protein containing 41 cysteine residues. The positions of twenty disulfide bridges of the recombinant protein expressed from CHO cells were determined by the combination of unique enzymatic digestion of the protein and the LC/MS/MS analysis. In addition, it was also identified that an unpaired cysteine was highly cysteinylated. The protein is currently under development as a therapeutic protein for liver failure diseases.
