LA-ICP MS Imaging for Elucidating the Mechanisms of Platinum Agent-Induced Liver Injuries in the Pig
Oxaliplatin (L-OHP) is a platinum (Pt) anticancer agent and is one of the effective drugs for the treatment of advanced colorectal cancer. However, it may cause liver injury, including sinusoidal obstruction syndrome (SOS), which would be associated with increased rates of morbidity after liver surgery in patients with colorectal liver metastases. Unfortunately, the mechanism of SOS is not clearly understood. In this study, we succeeded in establishing the pig model of SOS using L-OHP-based chemotherapy. Furthermore, laser ablation ICP mass spectrometry (LA-ICP MS) allowed us to visualize the characteristic distribution of Pt on the pig liver tissues with quantitative information. Combined with the pathological findings on HE stainings and blood Pt concentrations, our findings would give a clue to better understanding of the mechanism of SOS.
