Changes in protein expression levels in different carbon sources and their effect on cell growth rate

In order to adapt to a variety of growing conditions, proteome expression profile is cooperatively modulated for each functional group. In our previous study, it has been shown that expression abundances of two protein functional groups, protein synthesis and glucose/ethanol metabolism, are divergent among two yeast species having different growth rate under a non-fermentative carbon source, glycerol. Translation capacity in cells is known to be positively correlated with cell growth and glucose/ethanol metabolism is not essential for using glycerol as an energy source. Thus, it is suggested that modulation and optimization of protein expression pattern and proteome resource allocation can improve cell growth potential.
We are in progress of design of genetically modulated Saccharomyces cerevisiae that has proteome expression profile of functional groups related to protein synthesis and glucose/ethanol metabolism that are similar to Kluyveromyces lactis having higher growth rate under glycerol as a carbon source. We would like to present the genetically modulated proteome expression profiles and their influences on cell growth for deletion mutant for glycolysis and ethanol metabolism-related genes and for strains overexpressing transcriptional factor that control expression of genes encoding ribosomal proteins and translational elongation factors.
