Analysis of volatile compounds in mouth for oral diagnosis

We tried several sampling methods for volatile organic compounds (VOCs) in mouth by using solid phase micro extraction (SPME)-GCMS. The human volatilome is known as one of omics research. These compounds would be targets for oral diagnosis with rapid and easy analysis. In our previous study, we found 19 compounds in gingival crevicular fluid (GCF) as biomarker candidates for periodontal disease. However, these compounds are non-volatile such as amino acids, saccharides, organic acids etc. To establish rapid analysis for on-site diagnosis, we need to establish very fast analysis method including sample preparation, analyzing time, data reduction. In this study, we used SPME method for sampling of volatile compounds. This would be great advantage to analyze rapidly because we can remove pretreatment like TMS derivatization etc. We also tried to use microcrystallinewax-contained polymer (gum) for VOC metabolites collection and accumulation. This polymer was developed to collect stimulated saliva for medical analysis.
