Investigation of the Trap-column Suitable for C18 Modified Long Monolithic Silica Capillary Column

We investigated the trap column for long C18 monolithic silica capillary column. Preferable aspects of trap column are (1) lower retention than separation column, (2) low sample loss, (3) high sample load. To investigate the trap-column properties, monolithic silica capillaries were modified with C12 and C16 silylating agents, and evaluated the retention behavior with peptide mixture in gradient elution condition. As a result, equally or higher retention behavior of peptides were observed for both of C12 and C16 modified capillary columns, than those for a commercially available C18 monolithic silica capillary column. The reason is expressed by the difference of ligand density for C18 and the other ligands. C18 column was modified with bulky silylating agent, so surface density of ligands was relatively low while silylating agents of C12 and C16 were small. This result indicates it is important not only ligand type selection but surface density control to establish the suitable trap and elute LC/MS system for proteomics research.
