Mass spectrometric analysis of multiple catechol amines in brain by pyrylium-based derivative reagent.

In this time, we investigates the localization of catechol amines such as L-DOPA, dopamine(DA), 3-MT in which engage physiological change of mice brain. Py-I can selectively and efficiently react with the primary amino group which has more than two carbon chain by the reaction of pyrylium salts with nucleophiles under alkaline conditions, thereby increasing the mass by 105.1. Py-I-labeled L-DOPA and DA showed m/z 302.2 and 258.1 increasing the mass by 105.1 from original mass, respectively. MSI showed the change of localization of target molecules. Interestingly, L-DOPA and DA localized at brain stem and ventral striatum.
