Elucidation of atrial cardiomyocyte differentiation in iPS cells using LC/MS/MS
Human iPS cell-derived cardiomyocytes are anticipated to be applied to drug discovery and evaluation of safety. However, currently used iPSC-derived cardiomyocytes are mixture of subtypes, atrial cardiomyocyte, ventricular cardiomyocyte, and pacemaker cells, and the heterogeneities are not consistent. The reproducible derivation of pure or stable populations of the cardiac subtypes depends on our proper understanding for the mechanism of cardiac differentiation. Here, we report a mass spectrometric approach for elucidation of cardiac differentiation based on proteome analysis of undifferentiated iPS cell, iPS cell-derived mesoderm, and iPS cell-derived atrial cardiomyocyte.
