Quantitative analysis of hindlimb unloading mouse serum using DIA method
The data-independent acquisition (DIA) method, which was acquiring MS/MS data of all fragment ions, is more effective than the data-dependent acquisition (DDA) to reproducible and rapid analysis. In order to quantitative analysis of the mouse serum proteome, we firstly constructed a spectral library containing 1,072 mouse proteins from mouse serum. Then, through the DIA method using our spectral library, a total of 845 proteins were identified in three abundant proteins depleted and non-fractionated mouse serum from the hindlimb unloading model mouse (n=8) and the control (n=8). Of these 388 proteins were quantifiable proteins and 99 proteins were more than 2-fold differentially expressed between the two groups (p<0.05, t-test).
