Establishment of comparative quantitative analysis technique for skeletal muscle proteome
In this study, we aimed to establish a technique for comparative quantitative analysis of the skeletal muscle proteome. Skeletal muscle extracts from a mouse hind limb were fractionated into soluble and insoluble fractions, and then analyzed by LC-MS/MS to identify the constituent proteins. This analysis identified 505 and 418 proteins in the soluble and insoluble fractions, respectively. A total of 674 non-redundant proteins were identified, approximately 2-fold more than in a non-fractionated sample. Furthermore, we used this technique to analyze a large number of skeletal muscle samples obtained from mice bred under various conditions, and integrated the MS data using quantitative analysis software. Approximately 1,200 skeletal muscle proteins were quantitatively identified with high reproducibility.
