Construction of Mass Spectrometry Imaging System Using Infrared Matrix-Assisted Laser Desorption Electrospray Ionization

Mass spectrometry imaging (MSI) is a powerful technique for visualizing molecular distributions within tissues. However, common MSI methods usually operate under vacuum conditions. This requires sample dehydration and complex pre-treatment, which can lead to the loss of volatile components. In contrast, infrared matrix-assisted laser desorption electrospray ionization (IR-MALDESI) allows for measurements under atmospheric pressure with minimal sample preparation. In this study, a setup for MSI using IR-MALDESI was constructed to improve spatial resolution. The relationship between laser energy density and ion signal intensity was evaluated to find the condition for stable signal detection.
