Deep and Unbiased Plasma Proteomics at Scale with LC-MS

(Seer)
oMax Kosok

Mass spectrometry based proteomics is rapidly progressing, yet generating data that is both deep and scalable remains a persistent challenge. Many existing approaches rely on predefined targets or face practical limitations when applied to large sample sets, making it difficult to achieve broad, unbiased proteome interrogation at scale.

In this seminar, we will introduce Proteograph ONE, an automated workflow that integrates nanoparticle-based enrichment with LC-MS to enable comprehensive profiling of complex biofluids at peptide-level resolution. This approach extends measurable proteome coverage across a wide dynamic range, improving depth of coverage by 6-11x compared to traditional sample preparation, without modification to MS workflows.

Designed for high-throughput operation, the workflow standardizes sample preparation and supports reproducible performance across large cohorts. Its species-agnostic nature and compatibility with established mass spectrometry platforms allow for straightforward adoption.

Based on published case studies across diverse research areas, we will demonstrate how Proteograph ONE enables efficient scaling of proteomic studies while maintaining depth and delivering rich, unbiased datasets suited for discovery-driven research.
