Development of Intracellular Pharmacokinetic Evaluation Method Using a Single-cell Nano-delivery System

In this study, we developed a method for the direct intracellular injection of drugs using a single-cell nano-delivery system. We also established a single-cell mass spectrometry approach to analyze the injected drugs and their metabolites within individual cells. A mixture of amiodarone and propidium iodide (PI) was injected into HepG2 cells under microscopic observation using the single-cell nano-delivery system. The injected cells survived for more than 24 hours, indicating that the injection procedure exerted minimal effects on cell viability. After 24 hours of incubation, cells exhibiting PI-derived fluorescence were collected using a single-cell sampling system. As a result of mass spectrometry analysis of these cell samples, injected amiodarone and its metabolite, N-desethylamiodarone peaks were detected. These results show that the developed methods enable the direct intracellular delivery of drugs and subsequent metabolic analysis at the single-cell level.
